Australia had a relatively disappointing Olympic campaign in Rio 2016, with the swimmers not winning as many gold as was expected. Australia finished in 10th place on the medal tally (or 8th on ranking based on total medals won), with 29 medals (8 gold, 11 silver, and 10 bronze). Australia sent a team of 421, who competed in 26 sports.

Trivia
- Reigning Olympic champion in the 100m hurdles, Sally Pearson, withdrew from the Games due to a hamstring injury,
- Kitty Chiller, who competed as a modern pentathlete at the 2000 Summer Olympics in Sydney, was selected as the team's Chef de Mission, the first female to hold the role for Australia.
- Modern pentathlete Chloe Esposito entered the final leg of the women's modern pentathlon in seventh position, and finished in first, using her exceptional shooting and running skills.
- Cyclist Anna Meares competed at her fourth straight Olympic Games.
- Australia entered one artistic gymnast into the Olympic competition, failing to send any of the all-around teams for the first time since 1988.
- This was Australia's lowest medal tally and lowest rank since the 1992 Olympics in Barcelona where Australia also ranked tenth but only won 27 medals
